Conner had his arm in a sling after leaving the team's win over the Dolphins on Monday late in the fourth quarter. He appeared to land awkwardly on his shoulder on a tackle just before the two-minute warning, finishing with 145 yards rushing and a touchdown.
He has 380 yards and four scores on 97 carries through seven games this season.
Steelers coach Mike Tomlin told reporters Conner was being evaluated for an injury to the AC joint in his shoulder and he didn't practice all week.
Second-year running back Jaylen Samuels, who underwent knee surgery last month, took first-team reps in Conner's place and will likely be the starter against Indianapolis.
"Jaylen Samuels' availability is right on time," Tomlin said.
The Steelers (3-4) host the Colts (5-2) at 1 p.m. ET.
