Kvitova, the tournament's 32nd-seed, is a two-time Wimbledon champion but had no answers for a fired up Saville.
Australia's other remaining singles hopes Jason Kubler and Ajla Tomljanovic both fell in the second round but didn't go without a fight.
Doubles action also got underway on Day 4 with a number of Aussies including Thanasi Kokkinakis managing to progress to the next round.

Australian results at the French Open on Day 3
Time/Date (AEST) | Match | Score | Court | Event |
---|---|---|---|---|
11pm, May 24 | Hugo Gaston [FRA] v Alex de Minaur [AUS] | 4-6 6-2 6-3 0-6 7-6(4) | Suzanne Lenglen | Men's singles - First round |
10.10pm, May 24 | Ivan Dodig/Austin Krajicek v Filip Polasek/John Peers* | 6-4 7-6(2) | 5 | Men's doubles - First round |
In one of the biggest upsets of the day, Australian 19th seed Alex de Minaur was eliminated in the first round by France's Hugo Gaston 4-6 6-2 6-3 0-6 7-6(4), in a contest lasting almost four hours.
De Minaur struggled with the unpredictability of the 74th-ranked Gaston, even going behind in the first set, before he battled back to clinch it.
Gaston then drew on the boisterous French crowd - which De Minaur criticised post-match - to win the next two sets, before losing concentration to be bageled in the fourth.
However, the 21-year-old Frenchman outlasted De Minaur in a tiebreak in a fifth to move into a second round match with Pedro Cachin.
The Australian was disappointed with the crowd's behaviour, zeroing in on some particular things he didn't like about the atmosphere.
“I think there’s a difference between a great atmosphere and supporting your fellow countryman, which is completely fine and it’s great,” De Minaur said.
“For him, it was an amazing atmosphere. He enjoyed every second of it, but there’s a line.
“When I’m getting told things by people in the crowd, making eye contact with me after I hit a double fault, I think there’s a certain line that needs to be kind of looked at.
“Good for him for playing a great match in front of his home crowd and being able to feed off that, and having a moment that I’m sure he won’t forget.”

11.30pm, May 23 | Sebastian Korda [USA] v John Millman [AUS] | 6-1 7-5 7-6(6) | 6 | Men's singles - First round |
The second day of action at Roland-Garros began in brilliant fashion for Australians as Daria Saville swept past her Greek opponent Valentini Grammatikopoulou in straight sets and lost just three games in the process.
Australia's other remaining woman in the singles also pulled of an impressive victory as Ajla Tomljanovic knocked out fifth-seed Anett Kontaveit.
There was no such success for Australia's men's singles hopes as Jordan Thompson had no answers for clay king Rafael Nadal and James Duckworth failed to take a set off Swede Mikael Ymer.
John Millman's game against American Sebastian Korda was suspended after a single game due to bad weather.

Elsewhere, Thanasi Kokkinakis put up a good fight against Albert Ramos-Vinolas and came close to forcing their match to five sets.
